Analysis of Allantoin using HILIC HPLC
January 5, 2024
Introduction
Allantoin is a urea hydantoin that has an anti-inflammatory effect as well as acting as cellular activator and therefore commonly used in pharmaceutical and cosmetic products. Allantoin is highly hydrophilic, and it is recommended that a column is used that works using a separation mechanism based on hydrophilic interaction chromatography (HILIC). The analysis of commercial eyedrops were measured as reported in this note.
Experimental
Instrument
Pump | PU-2080 |
Vacuum degasser | DG-2080-53 |
Column Oven | CO-2060 |
Autosampler | AS-2057 |
Detector (UV-Visible) | UV-2075 |
Conditions
Column | FinePak SIL NH2-5 4.6mm x 250mm 5um |
Mobile phase composition | Acetonitrile/Water 80:20 |
Flow rate | 1mL/min |
Column temperature | 40 deg C |
Injection volume | 10uL |
Wavelength | 210nm |
Standard sample | Allantoin 5ug/mL in mobile phase |
Preparation
Dilute the eye-drops in mobile phase 10 times by volume, then filter using a 0.45um membrane filter.
