Abstract

The pulsed laser induced transient grating method is a powerful technique for measuring the diffusion coefficient (D) sensitively with a high time-resolution. For expanding the applicability of this method, a labeling method with a spiropyran derivate is demonstrated. The signals upon photoexcitation of the labeled proteins were strong and it was simple to determine D accurately. D and the circular dichroism spectra at various concentrations of a denaturant were measured and it was found that D changes before the circular dichroism spectrum change at a low concentration region, indicating that D is a sensitive probe for monitoring protein conformation.
